Description: Mouse metalloendopeptidase OMA1, mitochondrial (OMA1) ELISA kit (31.2-1000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of OMA1 in mice samples with a range of 31.2-1000 pg/mL and a minimum detectable dose (sensitivity) of 5.0 pg/mL. The kit is highly sensitive and easy to use.

Assay Principle: In this assay, purified mouse OMA1 antibody is used to coat microelisa strip plate wells to create a solid-phase antibody. OMA1 standards or samples and an HRP-labeled OMA1 antibody are added to the wells to form an antibody-antigen-antibody-enzyme complex. After thorough washing, the TMB substrate solution is added. The substrate turns blue under HRP catalysis and shifts to yellow upon the addition of a sulphuric acid stop solution. The color change is measured spectrophotometrically at 450 nm. The concentration of OMA1 in the samples is determined by comparing the OD of the samples to the standard curve.

Background: OMA1 is a mitochondrial inner membrane protease primarily functioning to hydrolyze the mitochondrial dynamics protein OPA1, thereby regulating mitochondrial fusion and the integrity of the mitochondrial intermembrane space. Under conditions of nutritional excess or metabolic stress, OMA1 is activated, leading to impaired mitochondrial fusion associated with insulin resistance and β-cell dysfunction. OMA1 activity represents a critical cellular response to metabolic stress and determines cellular fate (survival or apoptosis).
