Description: Mouse glucocorticoid receptor-beta (GR-β) ELISA kit is an ELISA-based in vitro research tool designed specifically for the quantitative detection of GR-β in mouse samples with a minimum detectable dose (sensitivity) of 0.1 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: The GR-β ELISA kit applies a competitive enzyme immunoassay technique utilizing a polyclonal anti-GR-β antibody and a GR-β-HRP conjugate. Assay samples and buffer are incubated together with the HRP conjugate in the pre-coated microplate for one hour. Because the number of binding sites is limited, higher concentrations of sample GR-β result in fewer sites available for the HRP conjugate. After washing the wells five times, an HRP substrate is added to develop color. The reaction is terminated with a stop solution, and the intensity is measured at 450 nm. The color intensity is inversely proportional to the target concentration, and sample values are calculated from the O.D. standard curve.
Background: GR-β is a splice isoform of the glucocorticoid receptor (GR). Unlike the physiologically active GR-α we are familiar with, GR-β cannot bind glucocorticoids and thus cannot mediate the classical glucocorticoid signaling pathway. Its primary function is believed to be the negative regulation of GR-α activity. It can form heterodimers with GR-α, preventing GR-α from entering the cell nucleus and binding to DNA, thereby inhibiting GR-α-mediated gene transcription. Consequently, GR-β is regarded as an endogenous inhibitor or antagonist of glucocorticoid action. Changes in GR-β levels may be associated with diseases such as obesity.
