Description:
Monkey 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (PFKFB3) ELISA kit-Quantitative competitive is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PFKFB3 in monkey samples with a minimum detectable dose (sensitivity) of 0.1 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The PFKFB3 ELISA kit utilizes a competitive enzyme immunoassay technique, employing a polyclonal anti-PFKFB3 antibody and a PFKFB3-HRP conjugate. Samples and assay buffers are co-incubated with the PFKFB3-HRP conjugate in the pre-coated plate for one hour. During this incubation, endogenous PFKFB3 from the samples competes with the PFKFB3-HRP conjugate for a limited number of antibody binding sites. After the incubation period, the wells are decanted and washed five times to remove unbound material. The wells are then incubated with an HRP substrate, forming a blue product that shifts to yellow upon the addition of a stop solution. The intensity of the color, measured at 450 nm, is inversely proportional to the PFKFB3 concentration. A standard curve is plotted to relate the optical density (O.D.) to the concentration of the standards, and the PFKFB3 concentration in each sample is interpolated from this curve.
Background:
PFKFB3 is a key bifunctional enzyme responsible for both the synthesis and degradation of fructose-2,6-bisphosphate (the most potent allosteric activator of the glycolytic rate-limiting enzyme PFK-1). Consequently, its activity is one of the primary determinants of cellular glycolytic flux. Given its pivotal role in glycolysis, PFKFB3 plays a significant role in diseases such as obesity.
