Summary:
INSR encodes an insulin receptor belonging to the receptor tyrosine kinase family. After proteolytic processing, the precursor protein forms a heterotetrameric receptor composed of two extracellular alpha subunits and two transmembrane beta subunits. Ligand binding activates downstream insulin signaling pathways that regulate glucose homeostasis as well as lipid, carbohydrate, and protein metabolism. Pathogenic variants in INSR are responsible for several inherited severe insulin resistance syndromes, including type A insulin resistance, Donohue syndrome, and Rabson–Mendenhall syndrome. Multiple transcript variants arise from alternative splicing.
