Summary:
The INS gene governs the production of Insulin, the core peptide hormone controlling carbohydrate and lipid metabolism. The gene product, Proinsulin, undergoes two precise post-translational cleavages to yield the mature, bioactive Insulin (A and B chains linked by disulfide bonds) and the C-peptide. The integrity of this hormone is vital, as its binding to the INSR triggers the systemic mechanism for glucose uptake. Genetic defects in INS can cause a spectrum of disorders, including permanent neonatal diabetes, different types of Maturity-Onset Diabetes of the Young (MODY) type 10, and conditions defined by excess immature hormone (hyperproinsulinemia).
