Description: Human tartrate resistant acid phosphatase 5b (TRACP 5b) ELISA kit (0.312-20 mIU/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of TRACP 5b in human samples with a range of 0.312-20 mIU/mL and a minimum detectable dose (sensitivity) of <0.06 mIU/mL. The kit is highly sensitive and easy to use.
Assay Principle: This kit features a microplate pre-coated with an anti-human TRACP-5b monoclonal antibody. The detection antibody provided is a biotinylated polyclonal antibody. Samples and biotinylated antibodies are added to the plate wells and cleared of unbound material with PBS or TBS. Avidin-peroxidase conjugates are then introduced. Following a thorough wash, TMB substrate is added to the wells. The enzyme reacts with the substrate to produce a blue complex, which shifts to yellow after the stop solution (color reagent C) is applied. The resulting color intensity is positively correlated with the concentration of the analyte in the sample.
Background: TRACP 5B, a metallophosphoesterase located at 19p13.2, encodes a tartrate-resistant acid phosphatase (EC 3.1.3.2) essential for bone resorption and the negative regulation of inflammatory cytokines, including TNF and interleukin-1 beta. By dephosphorylating osteopontin and bone sialoprotein, it maintains skeletal integrity and immune homeostasis; consequently, inactivating mutations lead to SPENCDI, while its elevated expression serves as a biomarker for various leukemias and Gaucher disease. In the context of metabolic health, TRACP 5B is increasingly recognized for its role in obesity-driven chronic inflammation, as its regulation of osteopontin signaling and macrophage activity directly influences the adipose tissue remodeling and systemic insulin resistance associated with excessive weight gain.
