Description:
Human prothrombin ELISA kit (0.5-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of prothrombin in human samples with a range of 0.5-10 ng/mL and a minimum detectable dose (sensitivity) of 0.1 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The prothrombin ELISA kit employs a quantitative sandwich enzyme immunoassay technique. The microtiter plate is pre-immobilized with a monoclonal antibody specific for prothrombin. Standards or samples are introduced into the wells, where any prothrombin present binds to the capture antibody. To quantify the target, a standardized preparation of HRP-conjugated polyclonal antibody specific for prothrombin is added to each well to "sandwich" the immobilized prothrombin. After incubation, the microtiter plate is thoroughly washed to remove all unbound components. Subsequently, substrate solutions are added, allowing the enzyme and substrate to react. Color development occurs exclusively in wells containing the prothrombin-antibody complex. The reaction is terminated with a sulphuric acid solution, and the absorbance is measured at 450 nm. A standard curve is plotted to relate the color intensity (O.D.) to the standard concentrations, from which the prothrombin concentration in each sample is interpolated.
