Description:
Human pro-melanin-concentrating hormone (PMCH) ELISA kit (15.6-1000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PMCH in human samples with a range of 15.6-1000 pg/mL and a minimum detectable dose (sensitivity) of <3.9 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This assay employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for PMCH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells, ensuring any PMCH present is captured by the immobilized antibody. After removing unbound substances through a washing step, a biotin-conjugated antibody specific for PMCH is added to the wells. Following a subsequent wash, avidin-conjugated HRP is added. After a final wash to remove any unbound avidin-enzyme reagent, a substrate solution is introduced, and color develops in direct proportion to the amount of PMCH bound in the initial step. The enzymatic reaction is terminated, and the intensity of the color is measured spectrophotometrically.

PMCH is a neuropeptide precursor synthesized in hypothalamic neurons, which is cleaved to generate melanocyte-concentrating hormone (MCH). MCH serves as a key neurotransmitter/neurotransmitter modulator involved in diverse neuronal functions, particularly regulating goal-directed behavior and overall arousal. PMCH and its derivative MCH represent critical targets for investigating the pathophysiology of obesity and the mechanisms governing appetite regulation.
