Description:
Human neuroendocrine secretory protein 55 (NESP55) ELISA kit is an ELISA-based in vitro research tool designed specifically for the quantitative detection of NESP55 in human samples with a range of 0.5-10 ng/mL and a minimum detectable dose (sensitivity) of 0.1 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The NESP55 ELISA kit utilizes a competitive enzyme immunoassay technique featuring a monoclonal anti-NESP55 antibody and an NESP55-HRP conjugate. Samples and assay buffer are incubated with the NESP55-HRP conjugate in a pre-coated plate for one hour. During this period, endogenous NESP55 from the sample competes with the NESP55-HRP conjugate for a limited number of antibody binding sites. After the incubation, the wells are washed five times. Subsequently, an HRP substrate is added, forming a blue complex that turns yellow upon the addition of a stop solution. The absorbance is measured at 450 nm. The color intensity is inversely proportional to the NESP55 concentration. A standard curve is generated to relate the O.D. to standard concentrations, from which the NESP55 levels in each sample are interpolated.
Background:
NESP55 is a protein expressed in multiple neuroendocrine tissues, particularly in the pancreas, adrenal medulla, and hypothalamus. It belongs to a protein family known as NESP, whose members are genetically closely linked and whose expression is regulated by a specific epigenetic mechanism—imprinted genes. As an imprinted gene product, its expression is influenced by parental origin and may serve as a potential regulator of energy balance, obesity, and others.
