Description: Human fatty acid synthase (FAS) ELISA kit (2.5-50 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of FAS in human with a range of 2.5-50 ng/mL and a specificity of 0.1 ng/mL.
Assay Principle: A microtiter plate is pre-coated with a monoclonal antibody specific for FAS. Standards or samples are then added to the wells of the microtiter plate, and FAS will bind to the antibody-coated wells if present. To quantify the amount of FAS in the sample, a standard preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody specific for FAS is added to each well to “clamp” the FAS immobilized on the microtiter plate. The microtiter plate is incubated, and the wells are then thoroughly washed to remove unbound components. A substrate solution is then added to each well. The enzyme (HRP) and substrate will react during the brief incubation period. Only wells containing FAS and enzyme-conjugated antibody will show a color change. Addition of sulfuric acid solution terminates the enzyme-substrate reaction, and the color change is measured spectrophotometrically at 450 nm. A standard curve is plotted between color intensity (O.D.) and standard concentration. The concentration of FAS in each sample is deduced from this standard curve.
Background: FAS is a multi-enzyme protein that catalyzes fatty acid synthesis. It is essential for the de novo synthesis of long-chain saturated fatty acids from acetyl-CoA, malonyl-CoA, and NADPH. Studies have found that FAS may be involved in obesity by regulating feeding behavior and energy homeostasis, potentially playing a role in body weight regulation and obesity development.
