Description: Human fatty-acid amide hydrolase (FAAH) ELISA kit (5.0-100 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of FAAH in human with a range of 5.0-100 ng/mL and a specificity of 1.0 ng/mL.
Assay Principle: The assay sample and buffer are incubated in a precoated plate with FAAH-HRP conjugate for one hour. At the end of the incubation period, the sample is decanted, and the wells are washed five times. The wells are then incubated with HRP enzyme substrate. The products of the enzyme-substrate reaction form a blue color complex. Finally, the reaction is stopped by the addition of a stop solution, which turns the solution yellow. The color intensity is measured spectrophotometrically at 450 nm on a microplate reader. The color intensity is inversely proportional to the FAAH concentration because FAAH in the sample competes with the FAAH-HRP conjugate for the binding site of the anti-FAAH antibody. Since there are a limited number of binding sites, as more sites are occupied by FAAH in the sample, there are fewer sites left to bind to the FAAH-HRP conjugate. A standard curve is plotted between color intensity (O.D.) and standard concentration. The concentration of FAAH in each sample is extrapolated from this standard curve.
Background: FAAH degrades bioactive fatty acid amides into corresponding acids, thereby terminating the signaling function of these molecules. It participates in neural signaling pathways involved in regulating feeding and is associated with obesity.
