Description: Rat insulin receptor substrate 1 (IRS-1) ELISA kit (0.313-20 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of IRS-1 in rat samples with a range of 0.313-20 ng/mL and a minimum detectable dose (sensitivity) of 0.188 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: This IRS1 ELISA kit is based on the sandwich-ELISA principle. The microplate has been pre-immobilized with an antibody specific to IRS1. Following the addition of standards or samples, a biotinylated detection antibody for IRS1 and an avidin-HRP conjugate are added in succession. After incubation and a thorough wash step to remove free components, a substrate solution is introduced for coloration. Only those wells containing the IRS1 sandwich complex will exhibit a color change. The reaction is terminated by an acidic stop solution, and the intensity is measured at 450 nm ± 2 nm. The resulting O.D. value is proportional to the concentration of IRS1 in the sample.
Background: IRS-1 is a key adaptor protein that plays a central role in insulin signaling. When insulin binds to its receptor on the cell surface, the receptor is activated and phosphorylates tyrosine residues on IRS-1. This process transforms IRS-1 into a signaling “relay station,” enabling it to bind with multiple downstream signaling proteins such as phosphoinositide 3-kinase (PI3K). IRS-1 transmits insulin signals from the cell membrane into the cell interior, thereby initiating a series of vital biological responses, including promoting glucose transport and glycogen synthesis. Studying the function and defects of IRS-1 is crucial for understanding the pathological processes underlying obesity.
