Description:
Mouse NAD-dependent protein deacetylase sirtuin-3; mitochondrial (SIRT3) ELISA kit-Quantitative sandwich is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT3 in mouse samples with a range of 0.312-20 ng/mL and a minimum detectable dose (sensitivity) of 0.156 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microtiter plate provided in this kit has been pre-coated with an antibody specific to the target antigen. Standards or samples are introduced into the designated microtiter plate wells along with a biotin-conjugated detection antibody preparation specific for the target antigen. Avidin-conjugated HRP is then added to each microplate well and incubated. After the TMB substrate solution is introduced, only those wells containing the complete sandwich complex will exhibit a color change. A sulphuric acid solution terminates the enzyme-substrate reaction, and the color change is measured spectrophotometrically at 450 nm ± 2 nm. The concentration of the target antigen is determined by mapping the sample O.D. values against the established standard curve.
Background:
Sirtuin 3 (SIRT3) is a soluble mitochondrial NAD+-dependent protein deacetylase that regulates the activity of key metabolic enzymes, including glutamate dehydrogenase and isocitrate dehydrogenase 2, thereby influencing the citric acid cycle, antioxidant regeneration, and cellular energy metabolism. SIRT3 is processed in the mitochondrial matrix to its active form and plays a central role in maintaining mitochondrial protein acetylation and metabolic homeostasis.
