Description:
Human beta-klotho (KLB) ELISA kit (62.5-4000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of KLB in human samples with a range of 62.5-4000 pg/mL and a minimum detectable dose (sensitivity) of 15.6 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This assay employs a quantitative sandwich enzyme immunoassay technique. A specific antibody for KLB has been pre-coated onto a microplate. Standards and samples are added to the wells, allowing any KLB present to be bound by the immobilized antibody. After removing unbound substances, a biotin-conjugated detection antibody specific for KLB is introduced. Following a wash, avidin-conjugated HRP is added. After a final wash to remove unbound reagents, a substrate solution is added; color develops in proportion to the amount of KLB captured in the initial step. The reaction is terminated, and the color intensity is measured spectrophotometrically.
Background:
KLB is a membrane protein serving as an essential co-receptor for fibroblast growth factor (FGF) signaling, specifically mediating the signaling pathways of FGF19 (intestinal origin), FGF21 (hepatic/adipose tissue origin), and FGF15. FGF21 is a crucial metabolic hormone that markedly improves insulin sensitivity, promotes energy expenditure, and reduces body weight, with KLB serving as the essential mediator for cellular reception of FGF21 signaling. Consequently, KLB function and expression are pivotal for investigating FGF21-mediated anti-obesity and anti-diabetic effects.
