Description: Human ATP-citrate synthase (ACLY) ELISA kit (1.563-100 IU/mL) is a ready-to-use ELISA kit for analyzing human ACLY levels with a detection range of 1.563-100 IU/mL and a sensitivity of 0.938 IU/mL.
Assay Principle: The kit is based on the sandwich ELISA technique. The anti-human ACLY antibody is pre-coated on a 96-well plate. The detection antibody is a biotin-conjugated anti-human ACLY antibody. Standards, test samples, and biotin-conjugated detection antibody are subsequently added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to visualize the HRP enzymatic reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of human ACLY sample captured in the plate. The concentration of human ACLY can be calculated by reading the absorbance of the external standard method at 450 nm using a microplate reader.
Background: ACLY is the major enzyme responsible for the synthesis of cytoplasmic acetyl-CoA in many tissues. The product it catalyzes, acetyl-CoA, is used in several important biosynthetic pathways, including cholesterogenesis and lipogenesis.
