Description:
Human apolipoprotein C-III (apo-CIII) ELISA kit (50-1000 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of apo-CIII in human with a range of 50-1000 ng/mL and a specificity of 1.0 ng/mL.
Assay Principle:
The assay sample and buffer are incubated in a precoated plate with the Apo-CIII-HRP conjugate for one hour. At the end of the incubation period, the sample is decanted, and the wells are washed five times. The wells are then incubated with the substrate for the HRP enzyme. The products of the reaction of the enzyme with the substrate form a blue color complex. Finally, the reaction is stopped by the addition of a stop solution, which turns the solution yellow. The color intensity is measured spectrophotometrically at 450 nm on a microplate reader. The color intensity is inversely proportional to the concentration of Apo-CIII because Apo-CIII and Apo-CIII-HRP conjugates in the sample compete for the binding site of the anti-Apo-CIII antibody. Since there are a limited number of binding sites, as more sites are occupied by Apo-CIII in the sample, fewer sites remain to bind the Apo-CIII-HRP conjugate. A standard curve is plotted to relate the color intensity (O.D.) to the concentration of the standard. The Apo-CIII concentration in each sample is extrapolated from this standard curve.
Background:
Apolipoprotein C-III is secreted by the liver and small intestine. It is a very low-density lipoprotein (VLDL) protein that inhibits lipoprotein lipase and hepatic lipase. Studies have found that it plays a key role in the metabolism of triglyceride-rich lipoproteins (TRLs).
