Description: Canine perilipin 1 (PLIN1) ELISA kit-Quantitative competitive is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PLIN1 in canine samples with a minimum detectable dose (sensitivity) of 0.1 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: The PLIN1 ELISA kit employs a competitive enzyme immunoassay technique. This method utilizes a monoclonal anti-PLIN1 antibody and a PLIN1-HRP conjugate. The test sample and buffer are co-incubated with the PLIN1-HRP conjugate in a pre-coated plate for sixty minutes. During this process, PLIN1 from the samples competes with the PLIN1-HRP conjugate for a finite number of antibody binding sites. After incubation, the wells are emptied and subjected to five wash cycles. Subsequently, an HRP enzyme substrate is added, forming a blue-colored complex. The reaction is halted with a stop solution, shifting the color to yellow. The absorbance is measured at 450 nm, where the color intensity is inversely proportional to the PLIN1 concentration. A standard curve is used to interpolate the sample results based on the recorded optical density (O.D.).
Background: PLIN1 is an adipocyte-specific lipid-binding protein that localizes to lipid droplets and plays a central role in regulating lipid storage and mobilization. By coating lipid droplets, PLIN1 protects stored triacylglycerols from basal hydrolysis while enabling controlled lipolysis in response to hormonal stimulation through phosphorylation by cAMP-dependent protein kinase. This dynamic regulation of lipid breakdown is essential for maintaining adipocyte lipid homeostasis. Dysregulation or mutation of PLIN1 impairs normal lipolytic control and fat storage, contributing to abnormal adipose tissue function and is closely associated with obesity and related metabolic disorders, including familial partial lipodystrophy.
