Summary: MC3R encodes the melanocortin-3 receptor, a G protein–coupled receptor responsive to melanocortin peptides such as melanocyte-stimulating hormone and adrenocorticotropic hormone. Unlike related receptors, MC3R is expressed in multiple tissues beyond classical endocrine targets. Functional studies demonstrate that loss of MC3R activity leads to increased adiposity in animal models, even in the presence of reduced food intake, indicating a role in energy expenditure and nutrient utilization. In humans, MC3R variants are linked to increased obesity risk.
