Description:
Rat uncoupling protein-1 (UCP1) ELISA kit (0.156-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of UCP1 in rat samples with a range of 0.156-10 ng/mL and a minimum detectable dose (sensitivity) of 0.091 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microtiter plate provided in this kit has been pre-coated with an antibody specific to UCP1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for UCP1, followed by the addition of avidin-conjugated HRP. After incubation, a TMB substrate solution is added to each well. Only those wells containing UCP1, biotin-conjugated antibody, and enzyme-conjugated avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution, and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of UCP1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uncoupling protein 1 (UCP1) is a mitochondrial transporter that mediates proton leakage across the inner mitochondrial membrane, uncoupling oxidative phosphorylation from ATP synthesis and dissipating energy as heat. UCP1 plays a key role in brown adipose tissue thermogenesis and energy expenditure, contributing to the regulation of whole-body metabolism. Impaired UCP1 function can reduce thermogenic capacity and promote energy storage, linking it to obesity and related metabolic disorders.
