Product Overview

Description: Rat plasminogen activator inhibitor-1 (PAI-1) ELISA kit-Quantitative sandwich is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PAI-1 in rat samples with a range of 0.625-40 ng/mL and a minimum detectable dose (sensitivity) of 0.319 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: The provided microtiter plate is pre-immobilized with an antibody specific to PAI. Standards or samples are added to the wells along with a biotin-conjugated polyclonal detection antibody for PAI, followed by the addition of avidin-conjugated HRP. After incubation and subsequent washing, a TMB substrate solution is introduced. Only wells containing the PAI-biotin-avidin complex will demonstrate a color change. The reaction is halted by a sulphuric acid solution, and the absorbance is recorded at 450 nm ± 2 nm. The PAI concentration is determined by comparing the O.D. of the samples to the standard curve.
Background: As the primary physiological inhibitor of tissue-type plasminogen activator and urokinase-type plasminogen activator, PAI-1 exerts its effects by suppressing fibrinolysis and proteolysis. Its elevation is associated not only with thrombosis and fibrosis but also with obesity and insulin resistance.
