Description:
Rat phosphoenolpyruvate carboxykinase (PEPCK) ELISA kit-Quantitative sandwich is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PEPCK in rat samples with a range of 15.625-1000 pg/mL and a minimum detectable dose (sensitivity) of 9.375 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This kit is based on sandwich ELISA technology. An anti-rat PEPCK antibody is pre-coated onto 96-well plates, and a biotin-conjugated anti-rat PEPCK antibody is employed as the detection antibody. Standards, test samples, and the biotinylated detection antibody are added to the wells sequentially, followed by a wash step with wash buffer. HRP-Streptavidin is then introduced, and unbound conjugates are removed through further washing. TMB substrates are used to visualize the HRP enzymatic reaction; the TMB is catalyzed by HRP to produce a blue product that shifts to yellow upon the addition of an acidic stop solution. The intensity of the yellow color is directly proportional to the amount of rat PEPCK captured in the plate. The optical density (O.D.) absorbance is measured at 450 nm in a microplate reader, allowing for the calculation of the rat PEPCK concentration.
Background:
PEPCK is a key rate-limiting enzyme in the gluconeogenesis pathway. Its overexpression and increased activity lead to insulin resistance and affect lipid metabolism and obesity.
