Background:
NR0B2 (also known as SHP) is an orphan nuclear receptor lacking a DNA-binding domain that inhibits the transcriptional activity of other nuclear receptors by binding to them. NR0B2's association with obesity stems from its broad inhibitory effects on glucose, lipid, and bile acid metabolism: SHP suppresses the expression of key metabolic genes, such as those involved in gluconeogenesis and fatty acid synthesis. In obesity and insulin resistance models, SHP activity serves as a pivotal regulator of hepatic glucose and lipid metabolic flux.
