Description: Rat hormone-sensitive lipase (HSL) ELISA kit (31.25-2000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of HSL in rat samples with a range of 31.25-2000 pg/mL and a minimum detectable dose (sensitivity) of 18.75 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle: This kit is based on sandwich enzyme-linked immunosorbent assay technology. An anti-HSL antibody is pre-immobilized onto 96-well microplates. A biotin-conjugated anti-HSL antibody is utilized as the detection agent. Standards, test samples, and the biotinylated detection antibody are added to the wells sequentially and washed with buffer. HRP-streptavidin is then introduced, and any unbound conjugates are removed with wash buffer. TMB substrates are used to visualize the HRP-catalyzed reaction, shifting from blue to yellow after the addition of an acidic stop solution. The density of the yellow signal is proportional to the HSL captured in the plate. The O.D. is read at 450 nm to calculate the final concentration.
Background: HSL is the key enzyme for fat breakdown. Belonging to the “GDXG” lipase family, this enzyme primarily hydrolyzes stored triacylglycerols into free fatty acids in adipose tissue and the heart. This process represents the rate-limiting step of lipolysis, controlling the overall speed of fat breakdown. HSL can be rapidly activated by catecholamine hormones (such as epinephrine) through a cyclic adenosine monophosphate (cAMP)-dependent phosphorylation process, while insulin regulates its dephosphorylation and inactivation. Due to its functional characteristics, HSL has become a central subject in obesity research, particularly in the regulation of fat storage and release.
