Description: Mouse apolipoprotein C-III (apo-CIII) ELISA kit (100-2500 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of apo-CIII in mouse with a range of 100-2500 ng/mL and a specificity of 1.0 ng/mL.

Assay Principle: The microtiter plate is pre-coated with a monoclonal antibody specific for Apo-CIII. Standards or samples are then added to the wells of the microtiter plate and will bind to the wells pre-coated with the antibody if Apo-CIII is present. To quantify the amount of Apo-CIII in the sample, a standard preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody specific for Apo-CIII is added to each well to “pinch” Apo-CIII immobilized on the microtiter plate. The microtiter plate is incubated, and then the wells are thoroughly washed to remove all unbound components. A substrate solution is then added to each well. The enzyme (HRP) and substrate will react during the short incubation period. Only wells containing Apo III and enzyme-bound antibodies will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is measured spectrophotometrically at 450 nm. A standard curve of color intensity (O.D.) versus concentration of the standard is plotted. The concentration of Apo-CIII in each sample is extrapolated from this standard curve.

Background: Apolipoprotein C-III is secreted by the liver and small intestine. It is a very low-density lipoprotein (VLDL) protein that inhibits lipoprotein lipase and hepatic lipase. Studies have found that it plays a key role in the metabolism of triglyceride-rich lipoproteins (TRLs).
