Description:
Human necdin ELISA kit (0.156-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of necdin in human samples with a range of 0.156-10 ng/mL and a minimum detectable dose (sensitivity) of 0.066 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The methodology employed in this kit is the sandwich enzyme immunoassay. The provided microtiter plate is pre-immobilized with an antibody specific to necdin. Standards or samples are added to the appropriate microtiter wells along with a biotin-conjugated antibody specific for necdin. Next, avidin-conjugated HRP is added to each microplate well and incubated. Upon the addition of TMB substrate solution, only wells containing the necdin, biotin-conjugated antibody, and enzyme-conjugated avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by a sulphuric acid solution, and the absorbance is recorded spectrophotometrically at 450 nm ± 10 nm. The concentration of necdin in the samples is determined by mapping the O.D. values against the standard curve.
Background:
Necdin is a protein located within neuronal cell nuclei and is considered a transcriptional repressor. It belongs to the genes within the critical region of Prader-Willi syndrome (PWS). PWS is a genetic disorder characterized by severe morbid obesity and hyperphagia, primarily attributed to the deletion or dysfunction of genes within the PWS region (including Necdin). This suggests Necdin participates in the normal suppression of satiety and food intake within the hypothalamus.
