Description: Human NAD-dependent protein deacetylase sirtuin-3; mitochondrial (SIRT3) ELISA kit (0.1-40 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT3 in human samples with a range of 0.1-40 ng/mL and a minimum detectable dose (sensitivity) of 0.051 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: This kit utilizes an ELISA based on the sandwich principle. The plate is pre-coated with a human SIRT3 antibody. SIRT3 present in the sample binds to the immobilized antibodies on the wells, followed by the addition of a biotinylated human SIRT3 detection antibody. Subsequently, streptavidin-HRP is introduced to bind the biotinylated antibody. After incubation, unbound streptavidin-HRP is removed during a washing step. Substrate solution is then added, and color develops in direct proportion to the amount of human SIRT3 present. An acidic stop solution terminates the reaction, and the absorbance is measured at 450 nm.
Background: Sirtuin 3 (SIRT3) is a soluble mitochondrial NAD+-dependent protein deacetylase that regulates the activity of key metabolic enzymes, including glutamate dehydrogenase and isocitrate dehydrogenase 2, thereby influencing the citric acid cycle, antioxidant regeneration, and cellular energy metabolism. SIRT3 is processed in the mitochondrial matrix to its active form and plays a central role in maintaining mitochondrial protein acetylation and metabolic homeostasis.
