Description:
Human lactase-phlorizin hydrolase (LPH) ELISA kit (0.156-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of lactase in human samples with a range of 0.156-10 ng/mL and a minimum detectable dose (sensitivity) of <0.05 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microtiter plate provided in this kit is pre-coated with an antibody specific to LPH. Standards or samples are introduced into the designated wells along with a biotin-conjugated detection antibody preparation specific for LPH. Next, avidin-conjugated HRP is added to each microplate well and incubated. Upon the addition of TMB substrate solution, only wells containing the LPH-biotin-avidin sandwich complex will exhibit a colorimetric change. A sulphuric acid solution terminates the reaction, and the absorbance is measured at 450 nm ± 10 nm. The concentration of LPH is determined by comparing the OD of the samples to the standard curve.
Background:
LPH is an enzyme located in the brush border of the small intestine, responsible for hydrolyzing lactose into glucose and galactose. It is an essential enzyme for lactose digestion. Genetic mutations in the lactase gene, associated with its activity persistence, determine lactose tolerance. This has potential implications for body weight and metabolic health.
