Description: Human cathepsin S (CTSS) ELISA kit (78.125-5000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of CTSS in human with a range of 78.125-5000 pg/mL and a specificity of 46.875 pg/mL.
Assay Principle: The capture antibody is pre-coated on a 96-well plate. A biotin-conjugated antibody is used as the detection antibody. Standards, assay samples, and biotin-conjugated detection antibodies are subsequently added to the wells and washed with washing solution. HRP-Streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to visualize the HRP enzymatic reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of target sample captured in the plate. The concentration of the target can be calculated by reading the absorbance at 450 nm with a microplate reader.
Background: CTSS is an important member of the cysteine protease family. As a potent mammalian elastase, it can degrade various extracellular components, including fibronectin, laminin, elastin, and collagen. CTSS is abnormally expressed in various disease states, such as obesity and atherosclerosis, which makes it a biomarker and a potential therapeutic target. Studies have found that CTSS inhibitors may be a possible strategy for treating obesity.
