Description:
Human aldo-keto reductase family 1 member C3 (AKR1C3) ELISA kit is an ELISA-based in vitro research tool designed specifically for the quantitative detection of AKR1C3 in human with a specificity of 1.0 pg/mL. The kit is highly specific and easy to use.
Assay Principle:
The assay sample and buffer are incubated in the precoated plate with the AKR1C3-HRP conjugate for one hour. At the end of the incubation period, the samples are decanted, and the wells are washed five times. The wells are then incubated with HRP enzyme substrate. The products of the enzyme-substrate reaction form a blue color complex. Finally, the reaction is stopped by the addition of a stop solution, which turns the solution yellow. The color intensity is measured spectrophotometrically at 450 nm on a microplate reader. The color intensity is inversely proportional to the concentration of AKR1C3 because the AKR1C3 and AKR1C3-HRP conjugates in the sample compete for the binding site of the anti-AKR1C3 antibody. Since the number of binding sites is limited, as more sites are occupied by AKR1C3 in the sample, fewer sites remain to bind the AKR1C3-HRP conjugate. A standard curve is plotted between color intensity (O.D.) and standard concentration. The concentration of AKR1C3 in each sample is derived from this standard curve.
Background:
AKR1C3 is a steroidogenic enzyme that catalyzes the conversion of aldehydes and ketones into alcohols and belongs to the aldo/keto reductase family. Studies have found that it may be associated with certain pathological processes.
