Description: Guinea pig brain cannabinoid receptor 1 (CB1) ELISA kit (0.156-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of CB1 in guinea pig with a range of 0.156-10 ng/mL and a specificity of 0.094 ng/mL.
Assay Principle: The micro ELISA plates provided in this kit are pre-coated with antibodies specific to CB1. Standards or samples are added to the appropriate micro ELISA plate wells and bound to the specific antibody. A biotinylated detection antibody specific for CB1 and avidin-horseradish peroxidase (HRP) conjugate are then added sequentially to each microplate well and incubated. The free components are washed away. Substrate solution is added to each well. Only wells containing CB1, biotinylated detection antibody, and avidin-HRP conjugate will appear blue. Upon the addition of sulfuric acid solution, the enzyme-substrate reaction terminates, and the color changes to yellow. Optical density (OD) is measured spectrophotometrically at 450 nm ± 2 nm. The OD value is proportional to the concentration of CB1. The concentration of CB1 in the sample is calculated by comparing the OD value of the sample with the standard curve.
Background: CB1 is a member of the cannabinoid receptor family and the G protein-coupled receptor (GPCR) superfamily. It is expressed at the highest levels in different brain regions, and is also present at lower levels in most other cell/tissue types, including adipose tissue. It is thought to activate multiple signaling pathways, regulating energy metabolism and cell survival/death. CB1 has become a drug target for treating obesity. CB1 inhibitors may be potential anti-obesity drugs.
