Description: Goat adipose triglyceride lipase (ATGL) ELISA kit is an ELISA-based in vitro research tool designed specifically for the quantitative detection of ATGL in goat. The kit is highly specific, sensitive, and easy to use.

Assay Principle: The microtiter plate is pre-coated with a monoclonal antibody specific to ATGL. Standards or samples are then added to the wells of the microtiter plate, and ATGL (if present) will bind to the wells pre-coated with the antibody. To quantify the amount of ATGL in the sample, a standard preparation of horseradish peroxidase (HRP)-conjugated specific ATGL polyclonal antibody is added to each well to “clamp” the ATGL immobilized on the microtiter plate. The microtiter plate is incubated, and then the wells are thoroughly washed to remove all unbound components. A substrate solution is then added to each well. The enzyme (HRP) and substrate will react during the short incubation period. Only wells containing ATGL and enzyme-bound antibody will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is measured spectrophotometrically at 450 nm. A standard curve is plotted between color intensity (O.D.) and concentration of the standard. The concentration of ATGL in each sample is derived from this standard curve.

Background: ATGL is a rate-limiting lipase in mammals, fruit flies, and yeast. It has very high substrate specificity for triacylglycerol (triglyceride) and catalyzes the first reaction of lipolysis, in which triacylglycerol is hydrolyzed into diacylglycerol (diglyceride). It is a key enzyme in maintaining the balance between lipid mobilization and lipid storage and affects various functions.
