Summary:
APOE encodes a major apolipoprotein that is essential for the normal catabolism (breakdown and clearance) of triglyceride-rich lipoproteins, notably chylomicron and VLDL remnants. APOE binds to specific receptors on the liver and peripheral cells, acting as the recognition marker necessary for cellular uptake. This gene resides in a cluster on chromosome 19 with other related apolipoproteins (C1 and C2). Specific mutations in APOE result in familial dysbetalipoproteinemia (Type III hyperlipoproteinemia), a disorder characterized by the impaired clearance of these remnants, leading to elevated plasma cholesterol and triglyceride levels.
