Background: TTC8 is a protein containing a tetrapeptide repeat domain that participates in ciliogenesis and vesicular transport. Mutations in the TTC8 gene cause Bardet-Biedl Syndrome (BBS). BBS is a hereditary ciliopathy characterized primarily by severe obesity. Dysfunction of TTC8 disrupts hypothalamic ciliary signaling, leading to appetite disorders and reduced energy expenditure.
