Background:
Patatin-like phospholipase domain-containing protein 3 (PNPLA3) is a multifunctional enzyme with both triacylglycerol lipase and acylglycerol O-acyltransferase activities, playing a key role in lipid metabolism and triacylglycerol turnover. Localized to membranes, PNPLA3 regulates lipid synthesis and catabolism in hepatocytes, and genetic variations in this protein are associated with susceptibility to non-alcoholic fatty liver disease (NAFLD) and related metabolic alterations.
