Description:
Mouse NAD-dependent protein deacetylase sirtuin-1 (SIRT1) ELISA kit (31.2-2000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT1 in mouse samples with a range of 31.2-2000 pg/mL and a minimum detectable dose (sensitivity) of 15.68 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microtiter plate provided in this kit is pre-coated with an antibody specific to SIRT1. Standards or samples are added to the microtiter wells with a biotin-conjugated polyclonal detection antibody specific for SIRT1. Subsequently, avidin-conjugated HRP is added to each well and incubated. A TMB substrate solution is then introduced; color development occurs only in wells containing the complete sirtuin-1 sandwich. The enzyme-substrate reaction is terminated by a sulphuric acid solution, and the absorbance is measured at 450 nm ± 2 nm. The concentration of SIRT1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Background:
Sirtuin 1 (SIRT1) is an NAD+-dependent protein deacetylase that links cellular energy status to transcriptional regulation, modulating processes such as metabolism, autophagy, apoptosis, and cell cycle control. SIRT1 deacetylates a variety of transcription factors and coregulators, influencing gene expression in liver, muscle, and adipose tissue, and participates in chromatin silencing through energy-dependent complexes. By sensing NAD+/NADH ratios, SIRT1 integrates metabolic signals and contributes to the regulation of glucose and lipid homeostasis, as well as cellular responses to stress and nutrient availability.
