Description:
Mouse 11 beta hydroxysteroid dehydrogenase type 1 (11-beta-HSD1) ELISA kit (0.312-20 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of 11-beta-HSD1 in mouse samples with a range of 0.312-20 ng/mL and a minimum detectable dose (sensitivity) of <0.115 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This kit utilizes the sandwich enzyme immunoassay technique. The microtiter plate is pre-coated with an antibody specific to 11-beta-HSD1. Standards or samples are added to the wells along with a biotin-conjugated detection antibody. Subsequently, avidin-HRP is added and incubated. Upon the addition of TMB substrate, a color change occurs only in wells containing the target complex. The reaction is terminated with sulphuric acid and measured at 450 nm ± 10 nm. The concentration of 11-beta-HSD1 is determined by comparing the O.D. of the samples to the standard curve.
Background:
11β-HSD1 is a key redox enzyme localized to the endoplasmic reticulum. It reversibly catalyzes the conversion of cortisol to the inactive metabolite cortisone. Although this enzyme exhibits bidirectional activity in vitro experiments, within living cells, it primarily converts inactive cortisone to active cortisol, thereby increasing local glucocorticoid concentrations. Obesity and type 2 diabetes are associated with abnormalities in glucocorticoid metabolism. The link between 11β-HSD1 and obesity represents a critical and extensively studied area of research.
