Description: Human proprotein convertase subtilisin/kexin type 1 (PCSK1) ELISA kit (0.312-20 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PCSK1 in human samples with a range of 0.312-20 ng/mL and a minimum detectable dose (sensitivity) of 0.101 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: The microtiter plate provided in this kit has been pre-coated with an antibody specific to PCSK1. Standards or samples are introduced into the designated microtiter plate wells along with a biotin-conjugated antibody preparation specific for PCSK1. Subsequently, Avidin-conjugated HRP is added to each microplate well and incubated. After the TMB substrate solution is added, only those wells containing the PCSK1-biotin-avidin sandwich complex will exhibit a colorimetric change. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution, and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 10 nm. The concentration of PCSK1 in the samples is then determined by comparing the O.D. of the samples to the established standard curve.
Background: Proprotein convertase subtilisin/kexin type 1 (PCSK1) is a serine endopeptidase that processes hormone and protein precursors at paired basic amino acid residues. Its substrates include POMC, renin, enkephalin, dynorphin, somatostatin, and insulin. Localized in the Golgi apparatus and secretory vesicles, PCSK1 plays a key role in peptide hormone maturation, proteolysis, and the regulation of insulin secretion.
