Description:
Human nicotinamide phosphoribosyltransferase (NAMPT) ELISA kit-Quantitative sandwich is an ELISA-based in vitro research tool designed specifically for the quantitative detection of NAMPT in human samples with a range of 0.313-20 ng/mL and a minimum detectable dose (sensitivity) of 0.188 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This kit is based on sandwich ELISA technology. An anti-human NAMPT antibody is pre-coated onto 96-well plates, and a biotin-conjugated anti-human NAMPT antibody serves as the detection agent. Standards, test samples, and the biotinylated detection antibody are added to the wells sequentially, followed by a wash step with wash buffer. HRP-streptavidin is then introduced, and unbound conjugates are removed through further washing. TMB substrates are employed to visualize the HRP enzymatic reaction; the TMB is catalyzed by HRP to produce a blue product that shifts to yellow upon the addition of an acidic stop solution. The intensity of the yellow color is directly proportional to the amount of human NAMPT captured in the plate. The O.D. absorbance is measured at 450 nm in a microplate reader, allowing for the calculation of the human NAMPT concentration.
Background:
NAMPT is the rate-limiting enzyme for nicotinamide adenine dinucleotide (NAD+) synthesis, catalyzing the conversion of nicotinamide to nicotinamide mononucleotide (NMN), which is then used to synthesize NAD+. NAMPT exists in two forms: the intracellular isoform (iNAMPT), which maintains intracellular NAD+ pools, and the secreted isoform (eNAMPT or Visfatin). The relevance of NAMPT to obesity research lies in NAD+ serving as a key cofactor for metabolic signaling and enzyme activity (e.g., Sirtuins). NAMPT activity serves as a central node linking nutritional status, energy metabolism, and aging. The secreted form of eNAMPT functions as an adipokine; studies have identified its abnormal expression in adipose tissue and serum of obese patients, suggesting potential roles in regulating insulin sensitivity and chronic inflammation.
