Description:
Human cathepsin A antibody (Cath Ab) ELISA kit is an ELISA-based in vitro research tool designed specifically for the quantitative detection of Cath Ab in human with a range of 2.5-50 U/mL and a specificity of 1.0 U/mL.
Assay Principle:
The microtiter plate has been pre-coated with a monoclonal antibody specific to Cath Ab. Standards or samples are then added to the microtiter plate wells, and Cath Ab (if present) will bind to the pre-coated antibody wells. To quantify the amount of Cath Ab in the sample, a standard preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody specific for Cath Ab is added to each well to “clamp” Cath Ab immobilized on the microtiter plate. The microtiter plate is incubated, and the wells are then thoroughly washed to remove all unbound components. The substrate solution is then added to each well. The enzyme (HRP) and substrate react during a short incubation period. Only wells containing Cath Ab and the enzyme-bound antibody will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution and the color change is measured spectrophotometrically at 450 nm. A standard curve of color intensity (O.D.) versus standard concentration is plotted. The concentration of Cath Ab in each sample is deduced from this standard curve.

CTSA is a lysosomal serine carboxypeptidase that provides protection for beta-galactosidase and neuraminidase. Cath Ab is a specific antibody designed to bind the CTSA.
