Customization of Transgenic Models
We inject exogenous genes into fertilized eggs or stem cells of animal models by microinjection and implant them into recipient animals to develop them into animals carrying the exogenous genes and capable of stable inheritance. By analyzing the correlation between the transgene and the phenotype, the function of the exogenous gene is analyzed, and studies on the molecular mechanism of obesity and anti-obesity therapies are conducted.
- Overexpression transgene model: We construct an expression vector with the target gene and promoter. Then the vector is injected into the fertilized eggs of animals using microinjection. We achieve broad and specific expression of the transgene by constructing different promoters, such as broad and tissue-specific promoters.
- RNA interference (RNAi) transgene model: Short hairpin RNA (shRNA) vectors are constructed and injected into fertilized eggs by microinjection. The interference effect of shRNAi is used to silence the expression of target genes.
- Bacterial artificial chromosome (BAC) transgenic model: BACs are effective tools for processing large segments of DNA and better explain the important functions of transgenes. BACs are either injected directly into prokaryotic microinjections or modified and injected into fertilized eggs in animal models to obtain transgenic animals. The model is used to study gene structure, spatiotemporal-specific gene expression, regulatory elements, etc.

Results: In this study, a transgenic mouse model expressing human serum amyloid A (hSAA) in adipocytes was constructed to investigate the role of SAA. The accuracy of the model has been verified by phenotypic and gene expression profiling of the mice by RT RCR, ELISA, etc. It was found that hSAA gene expression was unaffected by the high-fat diet, while plasma hSAA levels were increased compared to normal dietary feeding. The model is suitable for a detailed study of the metabolic role of SAA and its role in obesity.

What are the common techniques used for the construction of transgenic mouse models?
- DNA prokaryotic microinjection method: Exogenous genes are directly injected into fertilized eggs using microinjection so that the exogenous genes are integrated into the DNA of the recipients and develop into transgenic animals. This method is more widely used.
- Embryonic stem cell blastocyst microinjection: The exogenous gene is introduced into embryonic stem cells in vitro, and the transgenic embryonic stem cells are then injected into the blastocyst of the animal. This embryonic stem cell participates in the embryo formation of the host, forming chimeras until germline chimerism is achieved. Currently, the embryonic stem cell-mediated method is well-established in mice. Knockout and capture of genes are commonly used in this type of method.
What are the considerations when constructing transgenic animals?
Generally, the region of transgene expression depends on the promoter. Using a systemically expressed promoter will result in a systemically expressed transgenic animal. If a promoter for tissue-specific expression of the gene is chosen, tissue-specific expression of the transgenic mouse will be obtained. Due to the randomness of insertion of the transgene fragments, it is possible to insert into some repressive regions so that the gene is not expressed, or to insert into some enhancement regions so that the gene is highly expressed, and multicopy insertion usually occurs.
