Background:
VB12 is a water-soluble vitamin involved in metabolism. Foods that contain VB12 include poultry, fish, shellfish, liver, meat, dairy products, and eggs. VB12 is involved in cellular energy production and epigenetic regulatory processes, as well as being essential for single-carbon metabolism. Vitamin B12 deficiency is more common in obese individuals. Testing for VB12 provides useful information for obesity research.

Assay Principle:
VB12 supplies the growth of Lactobacillus delbrueckii, and the growth rate is associated with the volume of VB12 within the sample. The VB12 medium (Lactobacillus delbrueckii added), samples, or standards are taken and added to the microwell. Afterward, they are incubated at 37°C and protected from light. Lactobacillus delbrueckii will start growing until VB12 is consumed entirely. It takes 44-48 hours to complete incubation. A microplate reader is used to read the sample turbidity with a 610-630 nm or 540-550 nm filter. The volume of VB12 in the sample is determined by comparing the sample turbidity with the standards.
