Description:
Trolox equivalent antioxidant capacity (TEAC) assay kit is an antioxidant capacity assay kit that measures total antioxidant capacity based on the conversion of ABTS free radicals to ABTSW.
Background:
Oxidative stress is a physiological state of imbalance between reactive oxygen species (ROS) and antioxidant concentrations. Excessive ROS accumulation leads to DNA, protein, and cellular damage and is associated with a variety of diseases. Antioxidants include hydrophilic and lipophilic molecules used to metabolize ROS. These molecules may be transiently present in different tissues or cells. Due to their potentially harmful effects, excess ROS in cells must be removed promptly by various antioxidant defense mechanisms. Antioxidants usually neutralize free radicals through hydrogen atom transfer (HAT) or single electron transfer (SET) mechanisms. It is important to assess the antioxidant capacity of biological fluids, cells, and extracts.
Assay Principle:
In a 96-well microtiter plate, compare the sample to a known concentration of Trolox standard. Samples and standards are added to the wells of the microtiter plate, and the reaction is carried out for several minutes after the addition of the primed ABTS probe. The reaction is read at 405-415 nm using a standard 96-well spectrophotometric microplate reader. Antioxidant capacity is determined by comparing it to a standard.
