Background:
Bile acids usually consist of a 24-carbon steroid core and a side chain with a carboxyl group. The main metabolites of cholesterol in mammals are primary bile acids, including cholic acid and deoxycholic acid, which are secreted into the bile along with taurine or glycine. Primary bile acids undergo dehydroxylation to secondary bile acids, including deoxycholic acid and lithocholic acid. Disorders of bile acid metabolism are associated with liver disease, inflammatory bowel disease, nonalcoholic fatty liver disease, diabetes mellitus, and obesity.

Assay Principle:
Using S-NAD+ as the hydrogen acceptor, 3α-hydroxy steroid dehydrogenase catalyzes the dehydrogenation of bile acids to produce 3-ketosteroids, converting S-NAD+ to S-NADH. 3α-hydroxy steroid dehydrogenase catalyzes the generation of 3-ketosteroids to produce bile acids, with NADH used as the hydrogen donor. Through the cyclic reaction of the enzyme, S-NADH is continuously produced with a maximum absorption peak at 405 nm. The OD value is measured at 405 nm, and the change in absorbance is proportional to the concentration of bile acids.
