Background:
ADRB3, the β3-adrenergic receptor, regulates lipolysis, thermogenesis in brown adipose tissue, and vasodilation. β3-adrenergic receptor agonists have been found to have significant anti-obesity and anti-diabetic effects. ADRB3 is an attractive target in anti-obesity research.

Assay Principle:
The microtiter plate provided in this kit is pre-coated with an antibody specific for ADRB3. Standards or samples are then added to the appropriate microtiter plate wells along with a biotin-conjugated antibody preparation specific for ADRB3. Avidin conjugated to HRP is then added to each microplate well and incubated. Only wells containing ADRB3, biotin-conjugated antibody and enzyme-conjugated avidin will show a color change after addition of TMB substrate solution. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution and the color change is then measured spectrophotometrically at 450 nm ± 10 nm. The concentration of ADRB3 in the sample is then determined by comparing the OD of the sample to a standard curve.
