Background:
ADP is an important adipokine involved in the regulation of fat metabolism and insulin sensitivity, exhibiting antidiabetic, anti-atherosclerotic, and anti-inflammatory properties. It increases glucose utilization and fatty acid burning, stimulates AMPK phosphorylation and activation in liver and skeletal muscle, and may play a role in cell growth, angiogenesis, and tissue remodeling.

Assay Principle:
The kit is based on a sandwich ELISA technique. The anti-ADP/Acrp30 antibody is pre-coated on a 96-well plate. Biotin-conjugated anti-ADP/Acrp30 antibody is then used as the detection antibody. Standards, test samples, and biotin-conjugated detection antibody are subsequently added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to observe the HRP enzyme reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of ADP sample captured in the plate. The concentration of ADP can be calculated by reading the absorbance at 450nm with a microplate reader.
