Porcine adrenocorticotropic hormone (ACTH) ELISA kit (12.5-800 pg/mL) is a ready-to-use ELISA kit for analyzing porcine ACTH levels with a detection range of 12.5-800 pg/mL and a sensitivity of 7.5 pg/mL.
Background:
ACTH stimulates the adrenal glands to release cortisol. Defects in POMC may be associated with susceptibility to obesity (OBESITY). It is a condition characterized by an increase in body weight beyond the limitations of skeletal and physical requirements, as a result of excessive accumulation of body fat. Defects in POMC are the cause of pro-opiomelanocortinin deficiency (POMCD). Affected individuals present early-onset obesity, adrenal insufficiency, and red hair.

Assay Principle:
Pre-coat a 96-well plate with anti-porcine pro-adrenocorticotropic hormone antibody. Biotin-conjugated anti-porcine pro-adrenocorticotropic hormone antibody is used as the detection antibody. Standards, test samples, and biotin-conjugated detection antibodies are subsequently added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to observe the HRP enzyme reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of porcine ACTH sample captured in the plate. The concentration of porcine ACTH can be calculated by reading the absorbance of the external standard method at 450nm with a microplate reader.
