Description: Porcine acid phosphatase 5, tartrate resistant (ACP5) ELISA kit is a ready-to-use ELISA kit for analyzing porcine ACP5 levels with a sensitivity of 0.1 ng/mL. This kit has high sensitivity and excellent specificity.
Background: ACP5 is involved in the dephosphorylation of proteins. Its expression appears to increase in certain pathological states. Mice overexpressing ACP5 have been found to exhibit hyperplastic obesity. There are two phenotypes of this enzyme: 5a and 5b. TRAP 5a binds to preadipocytes and may exert cell number-regulating activity, inducing preadipocyte proliferation/differentiation. TRAP 5b may also induce the proliferation of certain cell types.

Assay Principle: The kit uses a competitive enzyme immunoassay technique utilizing a monoclonal anti-ACP5 antibody and an ACP5-HRP conjugate. The assay sample and buffer are incubated in a pre-coated plate with the ACP5-HRP conjugate for one hour. At the end of the incubation period, the wells are decanted and washed five times. The wells are then incubated with substrate for the HRP enzyme. The product of the reaction of the enzyme with the substrate forms a blue colored complex. Finally, the reaction is stopped by the addition of a stop solution, which turns the solution yellow. The color intensity is measured spectrophotometrically at 450 nm on a microplate reader. The color intensity is inversely proportional to the concentration of ACP5 because the ACP5 in the sample competes with the ACP5-HRP conjugate for the binding site of the anti-ACP5 antibody. Since the number of binding sites is limited, as more sites are occupied by ACP5 in the sample, fewer sites remain to bind the ACP5-HRP conjugate. A standard curve is plotted between color intensity (O.D.) and standard concentration. The concentration of ACP5 in each sample is derived from this standard curve.
