Description: Oxygen radical antioxidant capacity (ORAC) assay kit is an antioxidant capacity assay kit and provides sufficient reagents to perform the ORAC antioxidant capacity assay.
Background: Oxidative stress is a physiological state in which there is an imbalance between reactive oxygen species (ROS) and antioxidant concentrations. Under normal physiological conditions, ROS production by cells is counteracted by the action of cellular antioxidant enzymes and other redox molecules. However, excessive ROS accumulation can lead to DNA, protein, and cellular damage and is associated with a variety of diseases. Because of the potentially deleterious effects of excess ROS, they must be removed from the cell promptly through a variety of antioxidant defense mechanisms. It is important to assess the antioxidant capacity of biological fluids, cells, and extracts. ORAC assay is a classical tool to measure the antioxidant capacity of biomolecules in various samples.
Assay Principle: The assay is based on the oxidation of the fluorescent probe by peroxyl radicals through the process of hydrogen atom transfer (HAT). Peroxyl radicals are produced by a free radical initiator that quenches the fluorescent probe over time. Antioxidants in the sample prevent the peroxyl radicals from oxidizing the fluorescent probe until they are depleted. The remaining peroxyl radicals destroy the fluorescence of the fluorescent probe. The total peroxyl radical antioxidant activity in the sample is quantified from the fluorescence decay curve correlation.
