NADP+/NADPH assay kit (Colorimetric) is a metabolic assay kit for the rapid and sensitive detection of NADP+/NADPH in the range of 0.02-5.0 μmol/L.
Background:
NADP (nicotinamide adenine dinucleotide phosphate) is a coenzyme in many REDOX reactions, including NADP+ (oxidized) and NADPH (reduced) NADP+ is also involved in biosynthetic reactions such as lipid and nucleic acid synthesis. In animal cells, the oxidized phase of the pentose phosphate pathway is the most important source of NADPH.

Assay Principle:
Glucose 6-phosphate (G6P) is oxidized to 6-phosphate gluconolactone (6-PG) by glucose-6-phosphate dehydrogenase (G6PDH), and NADP+ is reduced to NADPH in this reaction. In the presence of 1-mPMS, NADPH transfers electrons to WST-8 to produce a yellow product, which has a characteristic absorption peak at 450 nm. Therefore, the total amount of NADP+ and NADPH can be quantified by measuring the OD value at 450 nm. After treating the sample and heating it for 30 minutes in a water bath at 60°C, the NADP+ of the sample is decomposed, leaving only NADPH, which reduces WST-8 to formazan. The amount of NADPH can be determined by measuring the OD value at 450 nm. The amount of NADP+ and the ratio of NADP+/NADPH in a sample can be determined from the total amount of NADP+ and NADPH obtained in the first two steps and the amount of NADPH alone. It should be noted that NAD+ and NADH do not influence the results.
