Assay Principle:
The microtiter plate provided in this kit is pre-coated with an antibody specific for ACADM. Standards or samples are then added to the appropriate microtiter plate wells along with a biotin-conjugated ACADM-specific polyclonal antibody preparation, and HRP-conjugated avidin is added to each microtiter plate well and incubated. TMB substrate solution is then added to each well. Only wells containing ACADM, biotin-conjugated antibody, and enzyme-conjugated avidin will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is measured spectrophotometrically at 450 nm ± 2 nm. The concentration of ACADM in the sample is then determined by comparing the OD of the sample to the standard curve.
